What a 483 response is, and why it matters

A 483 response is the firm’s written reply to the observations on a Form 483. FDA asks for it within 15 business days of the inspection’s close, and a timely, complete response is weighed before the agency decides whether to issue a warning letter.

A strong response takes each observation in turn: it acknowledges the finding, explains the investigation and root cause, lists corrective actions for the specific problem and preventive actions for the system behind it, and gives completion dates and evidence. Many firms follow up with progress updates until every commitment is closed.

Reading how other firms answered the same observation, especially ones that avoided a warning letter, is one of the fastest ways to write a response FDA will accept.

Questions

How long do you have to respond to a Form 483?

FDA asks for a written response within 15 business days of the end of the inspection. Responses received in that window are considered before FDA decides on further action.

What should a 483 response include?

For each observation: an acknowledgement, the investigation and root cause, corrective actions, preventive actions that address the wider system, completion dates and supporting evidence.

Is responding to a 483 mandatory?

A response is voluntary, but FDA strongly encourages one. Without a timely response, FDA has less reason not to escalate.

Why read other firms’ 483 responses?

They show how firms answered the same observations, what level of detail they gave and which commitments FDA accepted.
